Agenda balap dimulai pada Jumat, 16 April 2021 dengan sesi Free Practice 1 dan 2, untuk semua kelas. Sementara balapan berlangsung pada Minggu, 18 April 2021, mulai pukul 17.20 WIB.

Musim lalu MotoGP Portugal dimenangkan oleh pembalap Red Bull KTM Tech 3 – Miguel Oliveira. Pembalap tuan rumah itu menyelesaikan lomba dalam waktu 41 menit 48,163. Sementara posisi kedua diraih Jack Miller (Pramac Racing), sedangkan Franco Morbidelli (Petronas Yamaha SRT) finis di posisi ketiga.
